چکیده

The taxonomy of N2-fixing bacteria belonging to the genus Bradyrhizobium is still poorly refined. This paper presents an application of a method aiming the identification of possible new clusters within a Brazilian collection of 119 Bradyrhizobium strains showing phenotypic characteristics of B. japonicum and B. elkanii. The stability has been studied as a function of the number of restriction enzymes used in the RFLP-PCR analysis of three ribosomal regions. The method suggested here uses hierarchical clustering algorithms and Pearson correlation. Two clusters were clearly defined besides species B. japonicum and B. elkanii, indicating possible new species.

Endophytic Bradyrhizobium spp. isolates from sugarcane obtained through different culture strategies.

Brazilian sugarcane has been shown to obtain part of its nitrogen via biological nitrogen fixation (BNF). Recent reports, based on the culture independent sequencing of bacterial nifH complementary DNA (cDNA) from sugarcane tissues, have suggested that members of the Bradyrhizobium genus could play a role in sugarcane-associated BNF. Here we report on the isolation of Bradyrhizobium spp. isolat...

Phylogenetic diversity and cross-inoculation of indigenous isolated Bradyrhizobium from nodules of peanut in Liaoning province of China

Arachis hypogaea. L is a legume of economic importance, which is nodulated by Bradyrhizobium, a slow-growing bacteria. However there is no well characterization of this rhizobia in many areas of China. In the present study, cross-inoculation experiments were performed in cowpea and soybean. The isolated bacteria strains were characterized physiologyically, biochemically and id...

Application of representational difference analysis to identify genomic differences between Bradyrhizobium elkanii and B. Japonicum species

Bradyrhizobium elkanii is successfully used in the formulation of commercial inoculants and, together with B. japonicum, it fully supplies the plant nitrogen demands. Despite the similarity between B. japonicum and B. elkanii species, several works demonstrated genetic and physiological differences between them. In this work Representational Difference Analysis (RDA) was used for genomic compar...

Slow-growing and oligotrophic soil bacteria phylogenetically close to Bradyrhizobium japonicum

Eleven isolates of slow-growing oligotrophic bacteria from grassland soil were found to be closely related by partial 16S rRNA sequence similarity and many common taxonomic traits. Analysis of full 16S rRNA gene sequences of four representative isolates and Agromonas oligotrophica S58 indicated that they were more closely related to Bradyrhizobium japonicum, a symbiotic nitrogen-fixing bacteriu...

Proposal for combining Bradyrhizobium spp. (Aeschynomene indica) with Blastobacter denitrificans and to transfer Blastobacter denitrificans (Hirsch and Muller, 1985) to the genus Bradyrhizobium as Bradyrhizobium denitrificans (comb. nov.).

The symbiotic bradyrhizobia of Aeschynomene indica and the aquatic budding bacterium Blastobacter denitrificans have much in common and this study broadens the characters that are shared between the two. The 23S rRNA gene sequences of the bradyrhizobial isolates were most similar to each other and to the sequence of Bl. denitrificans. Evidence for the presence of photosynthetic genes in the gen...
